Product Description
Deoxyribonuclease I (DNase I), also known as Deoxyribonuclease I, is a nuclease found in a variety of cells and tissues. It is an endonuclease that cleaves the phosphodiester bond adjacent to the pyrimidine to produce a phosphate at the 5'end. Group, the 3'end of the polynucleotide is a hydroxyl group, the average digestion product is the smallest polytetranucleotide.
DNase I can catalyze many forms of DNA, such as single-stranded DNA, double-stranded DNA, and even chromatin (its cutting rate is affected by histones).
The best working range of DNase I is pH7~8.
The activity of DNase I depends on Ca2+ and can be activated by divalent metal ions such as Co 2+, Mn2+, and Zn2+. 5 mM Ca2+ can protect the enzyme from being hydrolyzed. In the presence of Mg2+, the enzyme can randomly recognize and cut any site on any strand of DNA; and in the presence of Mn2+, it can simultaneously recognize two strands of DNA and cut at almost the same site.
DNase I was first isolated from the pancreas, and the mammalian pancreas is also one of the most important sources of this enzyme.
DNase I is often used in molecular biology experiments to remove DNA in protein or RNA samples, or to introduce gaps in DNA to insert labeled bases into DNA.
The enzyme activity of this product is ≥2000U/mg protein.
